A disappointing game for the young Stags, who looked short of match practice in this local derby, played at Staveley, in what was their first outing of 2016.
Chesterfield looked like a completely different team to the one that John Dempster's side beat comfortably by three goals to one earlier in the season at Rainworth
The Spireites were ahead after just three minutes, when George Lynam forced the ball over the line from Derek Daley's second corner of the game.
Chesterfield had started the game at a high tempo and only a last ditch saving tackle from Kane Baldwin prevented Owen Darwent from getting a clear sight of goal.
Tom Jarrald came close to doubling the home sides lead, when he planted a header just over the bar from another Daley corner.
Mansfield's first chance of the game fell to Zayn Hakeem, but having beaten his marker the Stags attacker aimed his shot straight at Dylan Parker.
Kieran Harrison and Morgan Ratcliffe were doing their level best to keep a dominant Chesterfield attack at bay and Cain Smith had to help out at the back too, with a clearing header as the visitors withstood a lot of pressure from their local rivals.
Tom Marsham played Carlton Morrison in on goal, but the Spireites centre forward slipped as he cued up his shot, which allowed Sam Wilson to gather the ball.
The Stags only had one more clear chance in the first half, when Tyler Blake fed a sideways pass to the overlapping Tom Marriott, who struck his angled shot well but Parker saved.
Just before half time the home side had two more chances, but Morrison inadvertently blocked Owen Darwent's thumping effort from outside the box and Calum Greaves nudged a close range effort over the bar.
HT: Spireites 1 v Stags 0
Devante Reittie, who had been struggling to make any impression was substituted at half time and Tyler Johal came on in his place, but the Spireites looked strong in midfield and were making it difficult for Mansfield to create very much in the way of forward play.
The Stags did fare slightly better after the interval, but as Bloor, Blake, Hakeem and Marriott tried to undo their hosts, Chesterfield closed ranks at the back.
But while Mansfield tried to probe a way through for an elusive equaliser, the home side counter attacked at pace, but Daswell lobbed the ball over Wilson's goal from 12 yards out.
Johal's cross was scrambled away to Cameron Healey on the edge of the box whose first time shot was charged down by Lynam.
A Tom Marshall free kick deflected over the Stags defensive wall, but Wilson was alert to the danger.
Drew Gibson, on in place of Tom Marriott in the 63rd minute, powered forward and spread the ball wide to Baldwin, but Parkin got up well to punch the Mansfield full back's cross away.
The home side had to defend their goal now as the Stags picked up the pace, but their back four, and Lynam in particular, were in a determined mood today.
Harrison looked composed at the back for the visitors today and his distribution was excellent, but Mansfield weren't making good use of the ball in the final third.
Back at the other end, Daley crashed a 20 yard free kick against the right hand upright.
Chesterfield had the better of this afternoon's game, but they left it late to seal their win when Daswell netted from close range inside the final two minutes.
The Stags pushed forward in the closing stages, but two Teddy Bloor corner kicks came to nothing.
